Comparative Analysis of 26 Day Embryonic RNA (Carnegie 12, Source R318) Constructed cDNA and SAGE Libraries

To directly compare the frequencies of specific genes in each of these sets we chose 4 SAGE tags that were detected >10 times, 3 that were detected ~10 times and 3 that were detected 1-2 times per 50,000 sequenced tags. PCR primers were designed from the corresponding cDNA sequences and these were used to amplify each sequence from the 26 day embryonic RNA (Carnegie 12, source R318) constructed cDNA and SAGE libraries. Each of these probes was then separately hybridized to colony lifts from the C12 library.
